In continuation of the first funding period, we will measure the major endocannabinoids (eCBs) for the project research groups. In addition, we propose to extend our investigations “beyond CBs" by including additional eCB-related analytes and to develop extraction methodologies for new types of biological samples. We further intend to go "beyond the eCB system (ECS)” and to use lipidomic technologies for more comprehensive investigations of the interactions of the ECS with other lipid signaling systems and the general cellular lipid metabolism. The fast scan speed and high sensitivity of the AB SCIEX 5500 QTRAP, a triple-quadrupole linear ion trap hybrid instrument, have already been proven to be invaluable for eCB quantification by MRM (multiple-reaction-monitoring). Regarding "beyond ECS" analyses, we will combine the instrument's unique linear ion trap capabilities with information-dependent acquisition to establish different types of lipidomic workflows (e.g., targeted versus non-targeted approaches).
