Obesity-induced chronic inflammation is a major risk factor for non-communicable diseases. While the mechanism of chronic inflammation caused by over-supply of carbohydrates and fat is not fully understood, mouse models of numerous non-communicable diseases point to a common role of a single inflammatory key pathway, the NLRP3 inflammasome. This project aims to systematically delineate all molecular events leading to NLRP3 activation in human cells in response to metabolic conditions, cellular danger signals, and pathogen infections, using CRISPR-based functional genomics technologies.
