P6 proposes a complementary analysis in Drosophila and mouse models to investigate the mechanistic role of endocytosis. They will explore the mechanisms of endocytosis and cargo processing of Drosophila nephrin, and assemble a humanized slit diaphragm model by transgenesis of human slit diaphragm proteins to perform a molecular analysis of human slit diaphragm proteins in Drosophila. Lastly, endocytosis and dynamics of nephrin at the mammalian slit diaphragm will be studied utilizing injection of anti-nephrin antibodies in mice for live antibody labeling, as well as the generation and phenotypic analysis of a conditional knockout mouse with podocyte-specific deletion of Tbc1d8b.
