The Mannose Receptor (MR) is released as a soluble molecule (sMR) in the extracellular space. We have shown that increased sMR levels upon Western diet (WD) leads to macrophage activation, stimulation of weight gain, and metaflammation. We now aim to elucidate the mechanisms underlying MR shedding and its regulation upon WD challenge. We will further delineate the role of sMR in lipid metabolism in adipocytes and its role in microglia activation and stress vulnerability in the brain. Finally, we will investigate whether the impairment of MR-mediated macrophage activation can be used as therapeutic approach in metaflammation.
