This project focuses on the characterization of selected chloroplast proteins involved in cold acclimation and cold-induced degradation of envelope proteins. FAX1 degradation by RBL11 appears to be induced by post-translational modifications (PTMs), and this mechanism and its significance will be analyzed by reconstitution of both proteins (and modified variants) in vesicles, expression in E. coli and in transgenic plants. The proteins involved in the PTM, the importance of other uncharacterized envelope proteases for cold acclimation and candidate proteins from a plastidic artificial microRNA library screen will be further elucidated.
