The innate host defense system reacts quickly and adequately against invading pathogens. Thereby the immune response is supported by the release of the alarmin S100A8/A9 initiating a TLR4 dependent immune cell activation. Heterodimeric S100A8/A9 inactivates itself by calcium-dependent tetramer formation restricting the immune response locally. In the last funding period we will characterize the implications, if this regulatory mechanism cannot take place anymore in infectious mouse models and develop a nanobody/antibody based strategy to block or neutralize the activity of S100A8/A9 heterodimers.
