We aim to reconstruct the first representative dataset of rye highly-repetitive sequences in silico on a whole-genome scale. We will use a high-throughput 454 sequence survey for comparative analysis of the repeat composition with other grass species, for an improved analysis of large genomes. The sequence analysis of flow-sorted rye B chromosomes (Bs) will allow insights in the evolution of Bs. The chromosomal distribution of repeat families will be determined experimentally to establish chromosome specific makers. This analysis will provide new evidence for the generation of rapid genome changes in higher eukaryotes, with particular relevance to selfish chromosomes.
