The aim of this project is the generation, characterisation and provision of specific monoclonal antibodies, as needed in the majority of the projects of PROTRAIN. In particular, antibodies are wanted that recognize conformational epitopes and thus can discriminate between proteins prior to and after loading with prosthetic groups. In detail, this program comprises immunization of mice, monitoring of serum titers, isolation of spleen cells, fusion with precultivated myeloma cells, hybridoma selection, identifcation and characterisation of secreted antibodies by ELISA and immunoblotting, selection, cultivation and storage of productive clones, and finally the identification of antibody epitopes. A central facility with the required infrastructure is economic and will allow to place these tasks into expert hands. Therefore, this project is asking for the position to employ an experienced technical assistant who has been working very successfully in the field of generating epitope-specific antibodies for many years, and for the respective means for consumables.
