In vivo application of polySia-based materials in a rat model of peripheral nerve regeneration - Impact on morphological and functional regeneration

Final Report Abstract

After soluble polySia demonstrated no negative but rather regeneration promoting effects in the context of peripheral nerve repair, more solid polySia based materials were proposed to be delivered to the Institut für Neuroanatomie. The materials delivered were then analyzed for their applicability as biomaterials for peripheral nerve repair. Till the end of the funding period no promising candidate material could be identified. However, recently nanosilica particles coated with polySia have been integrated into collagen matrices which were then rolled to achieve nerve conduits (Institut für Anorganische Chemie). Further experiments will be performed in cooperation of the Institut für Anorganische Chemie and the Institut für Neuroanatomie. The study design includes short and long term studies as follows: 1) over a four week period the nerve regeneration distance across a 10mm nerve defect will be evaluated with histological methods in 3 groups A) pure collagen tubes, B) collagen tubes infiltrated with plain nanosilica particles and C) collagen tubes infiltrated with polySia coated nanosilica particles. In parallel functional recovery will be analysed in addition to nerve histomorphometry over a period of 12 weeks in the same groups plus a D) control group transplanted with an autologous nerve graft. In case group B) and or C) demonstrate good regenerative potential the study will be advanced by 13mm nerve gap repair and monitoring of functional and axonal regeneration over a maximum of 17 weeks. The study will elucidate if incorporation of nanosilica particles coated with polySia represent a nerve regeneration promoting building block of artifical nerve grafts. To our knowledge this will be the first study on the effects of polySia immobilized within nerve conduits.
