Project Description

The transporter associated with antigen processing (TAP) is a focal point in adaptive immunity against infected or malignantly transformed cells. This ABC transporter shuttles proteasomal degradation products into the endoplasmic reticulum for loading onto major histocompatibility complex class-I molecules (MHC-I). The peptide-loading complex (PLC) orchestrates the peptide translocation, editing, and transfer to MHC-I molecules. We aim at investigating the translocation mechanism, conformational states, and their dynamics of TAP, related ABC transporters and the PLC using integrative structural and functional approaches.
