Refined protocols for the co-translational insertion of cell-free synthesized G-protein coupled receptors into preformed nanodisc membranes and other environments revealed essential lipid dependencies of the beta-adrenergic and endothelin receptors for obtaining stable functional conformations. The molecular analysis of the receptors and their complexes with nanodiscs as well as their ligand recognition mechanisms in defined environments will now be approached by a number of complementary techniques. The potential of receptor/nanodisc complexes as antigens exposing folded domains for the isolation of specific antibodies will further be assessed.
