The confocal microscope is requested for current and future research projects for which currently no suitable equipment is available at the ZMBE, the place where zebrafish embryos are generated and manipulated. The microscope is requested for following fast dynamic processes at the sub-cellular level. Specifically, using this instrument, the turnover and mobility of membrane components and cytoskeletal elements could be determined during dynamic processes of cell migration using different experimental systems. The equipment would be particularly useful when studying zebrafish, A fish facility where this organism is reared and manipulated is located in the Institute. Equipment with the specifications required for rapid activation of fluorescent proteins and uncaging (requiring a UV laser), for other fast in vivo applications such as fluorescence recovery after photobteaching (FRAP) or fluorescence loss in photobleaching (FLIP) and in an upright configuration allowing easy manipulations is not available in the center. The long duration of experiments on live material make it essential to have dedicated equipment available right at the site where zebrafish embryos are produced and manipulated. The new equipment would further strengthen the local SFB initiatives, members of which could use the equipment with their own systems as well as when studying cellular dynamic process in zebrafish embryos.
