Folate metabolom studies and the effects of MTHFR mutation and folic acid supplements on folate forms: We established methods for quantifications of different forms of the vitamin folate in serum and whole blood. The methods are being used for clinical investigations. For example, we studied the effect of short term folic acid supplementation on folate form distributions and the appearance of free unmetabolized folic acid in blood. We further studied the folate forms in cord blood immediately after birth and the effect of maternal folic acid supplement usage on folate forms in cord blood. We also studied the effect of supplementing folic acid or the co-supplementation of B12 and B6 on the levels of free folic acid in blood. The interrelation between methylation, choline, phospholipids and the B-vitamins in Down syndrome, neurodegeneratlon and patients with diabetes: We established methods for measuring betaine, choline, acetylcholine, S-adenosylmethionine, S- adenosylhomocysteine, and over 20 species of the phospholipids. The methods were used and are being used for numerous clinical investigations. For example, we studied one-carbon metabolism and lipid metabolom in Down syndrome. People with down syndrome have an additional copy of the cystathionine beta synthase and superoxide dismutase. This causes severe changes in the methylation cycle and the phospholipid metabolism. We combined our assays with a mathematical model that has been developed in the USA and developed a therapeutic approach aimed at changing the metabolism in Down syndrome. Another study is currently running on lipid metabolom in multiple sclerosis, one classical disease where myelin is damaged. Because of the role of phospholipids in myelin formation, we propose that the lipidomic studies might help explaining the pathogenesis of the disease and open new therapeutic possibilities. The role of choline and betaine as methyl donors during pregnancy and prenatal life: We studied choline metabolites and the phospholipids in maternal and cord blood. The study aimed at finding factors that interfere with folate metabolism and save folate as a methyl donor during the prenatal life. Our studies on phospholipids in cord blood in relation to choline and folate metabolism will add more information on possible role for choline in prevention of birth defects.
