Project Description

Unlike the structures of proteins forming the clathrin and COPII coats, the structure of the COPI coat protein complex coatomer is unknown. We aim at elucidating the structure of individual coatomer isotypes by cryo-electron microscopy and single particle reconstruction. Further we plan to investigate by Cryo-EM individual steps of the coating process in a chemically characterized liposomal system, in which we add Arf1, p24 family proteins, nucleotides, and defined recombinant coatomer complexes obtained by expression in Sf9 cells.
