The project focuses on the combination of catalytic heme-binding peptides as model systems for peroxidases and peroxygenases and deep eutectic solvents (DES). For many enzymatic reactions, the use of DESs has been shown to improve process performance. However, the molecular effects and interactions between the DES and the enzymes that lead to these performance improvements are not yet understood. SP5 will provide fundamental information on the sequence-stability and sequence-activity relationships of heme-binding peptides and proteins such as peroxidases and peroxygenases in DESs to improve the predictability of these interactions by linking experimental and model-based work. The main working hypothesis of SP5 is that the use of short catalytically active heme-binding peptides with peroxidase activity can be used to elucidate these effects at the molecular level, focusing on the enzymes on the active side. To achieve these goals, existing preliminary work in the Holtmann group on DESs, peroxidases and peroxygenases as well as peptide synthesis will be combined with the expertise of the research unit to investigate these central questions in the field of DESs. The investigations will focus on peroxidases and peroxygenases as examples, since these enzymes are of great scientific and technical interest and fundamental preliminary work on the enzymatic catalysis of peroxidases and peroxygenases in DES is available.
