Project Description

Transregio 5 is an interdisciplinary research network focused on understanding the regulatory dimension imposed on genes by their chromatin organisation. The problem is addressed at various structural levels ranging from single nucleosomes and nucleosomal fibers to chromosomal domains and entire chromosomal territories employing well-developed experimental systems and selected model organisms. This integrated approach promises novel insights into basic principles and mechanisms governing central nuclear functions in eukaryotes including gene regulation, DNA replication and the repair of damaged DNA. Major goals of the programme are
-- to determine the factors and processes involved in the positioning and posttranslational modification of nucleosomes with regulatory function;
-- to assess the functional consequences of specific chromatin structures, considering interaction of non-histone regulators, chromatin folding and nuclear compartmentalisation;
-- to explore the principles by which enzymes that modify chromatin are targeted to defined gene loci, chromosomal domains and nuclear compartments;
-- to understand the principles which govern dynamic chromatin transitions and stable epigenetic memory.
