E3 ligases play an important role in the tumorigenesis of acute myeloid leukemia (AML) and in the resistance of AML to chemotherapy. Here, we propose to target two E3 ligases, RNF4 and TRIM24, using PROTACs that we will develop in the first phase of this CRC. We aim to establish a proof of concept for RNF4 and TRIM24 degradation as a novel therapeutic strategy in AML and to gain mechanistic insights into the cellular signaling network mediated by these E3 ligases using disease relevant cellular systems as well as in vivo models of AML.
