A novel strategy for the total synthesis of macrocyclic natural products will be established in the synthesis of antifungal agent 15G256ß. This includes the biomimetic synthesis of arenes by cyclisation of polycarbonyl compounds under very mild conditions as well as the construction of a macrolactone. This strategy will allow a new approach to numerous macrocyclic bioactive natural products bearing the 6-alkyl-2,4-dihydroxybenzoic unit. Furthermore two independent routes to the key building blocks for 15G256ß are suggested.
