The functionals role of AAA+ proteases as central components of the mitochondrial protein quality control network under normal and stress conditions

Molecular chaperones fulfill essential functions during cellular biogenesis, including biosynthesis and degradation of proteins. The mitochondrial matrix contains chaperones of the Hsp70, Hsp60 and Hsp100/Clp family. As the role of chaperones during protein translocation and folding is well characterized, only limited information is known about their involvement in degradation reactions. Aim of the project is the characterization of the functional cooperation of the mitochondrial chaperone network with the components the proteolysis system. The activity of the proteolytic system and its dependence on chaperone components shall be quantified under substrate saturating conditions. The spectrum of native substrate proteins of the mitochondrial degradation system will be analyzed in organello and in vivo to define the qualitative and quantitative substrate flow through the mitochondrial proteolysis system. The influence of chaperones and proteases on protein aggregation shall be determined. The characterization of the differential role of chaperone proteins during proteolysis will be the basis for the full understanding of the mitochondrial biogenesis.
