The measurement of distances between nuclear and electron spins by magnetic-resonance methods plays a fundamental role in structural biology. However, nuclear magnetic resonance (NMR) techniques typically suffer from three aspects in the context of large biomolecular complexes, namely low sensitivity, limited selectivity and the difficulty to obtain long-range distance information. This project aims to overcome these limitations by developing dynamic nuclear polarization (DNP)-based hyperpolarization approaches utilizing site-specific polarization transfer from an endogenous paramagnetic metal ion or introduced metal-chelate spin label to surrounding nuclei.
