This project aims to improve analytical methods to detect states in signaling networks that could predict drug sensitivity. A targeted phosphoproteome (PP) panel for the NB will be designed, and comprehensive catalogs of on- and off-target effects for drugs used in perturbation studies will be generated. A computerized tool will be developed to convert PP profiles into easily interpretable metabolic pathway activity values, and then applied to a representative cell line panel. Computer models will be used to hypothesize resistance mechanisms and synergies for combinatorial treatments.
