In the project, Henneke will explore molecular and cellular factors in macrophages and their progenitors underlying intestinal inflammation in chronic granulomatous disease (CGD). Two mouse models of CGD, as well as material from affected patients form the basis of investigations extending from bone-marrow to large intestine and involving e.g. single-cell tracing, high resolution tissue imaging and chronic infection models. Specifically, we will investigate the subset-specific role of NOX2 in monocytes and macrophages in spontaneous and inflammation-induced differentiation, the impact of microbial factors in macrophages modulating CGD colitis, and the origin and pathological contributions of non-conventional macrophages emerging in CGD colitis.
