Project C06 is aimed at studying dynamic endothelial cell-mural cell interactions during metastatic progression. Employing scRNAseq of vascular niche cells and proteomics of lung tissue, the effect of either genetic or pharmacological targeting of the TGFß pathway modifier LRG1 on mural cells and subsequently on the composition of the extracellular matrix will be assessed. Building on preliminary data, the temporal evolution of metastatic endothelial cells will be mapped following immunemodulatory interventions. Taken together, project C06 is expected to not only shed light on endothelial cell-stromal cell crosstalk, but also pave the way for mechanism-guided angiocrine therapy for limiting metastatic progression.
