A high-resolution tandem mass spectrometer with ultra high performance liquid chromatography coupling for the untargeted analysis of complex biological and synthetic samples is applied for. Therefore, a mass spectrometer with high resolution, mass accuracy, mass stability, scan rate and a broad dynamic range is requested. To enable identification of unknown and known compounds, the instrument should enable acquisition of the high resolution mass, as well as MS/MS-spectra. Thus, a tandem mass spectrometer with electronspray (ESI)-ionsource and a quadrupol- time-of-flight detector (Q-ToF) is applied for. The liquid chromatography system should include a binary high pressure pump, an autosampler with cooling system, and a diode array detector. Besides untargeted analytical questions, the instrument will be utilized for the biochemical characterization of novel enzymes, and the activity-screening of enzyme variants for protein engineering projects.
