This proposal aims to set-up a high-end cryo-electron microscopy (cryo-EM) facility that is equipped with instrumentation for the major current techniques of cryo-EM, namely single-particle analysis, cryo-electron tomography (cryo-ET) and electron diffraction as a university core facility. This will allow the University of Freiburg to be competitive in these seminal tech-niques of current structural and cellular biology. To realize this, a high-end 300 kV transmis-sion electron microscope with a fast-counting detector and an energy filter is necessary. For sample preparation in cryo-ET and further analysis, a cryo-focussed ion beam - scanning electron microscope in combination with a correlative light-and electron microscopy set-up is also required. Finally, as these instruments produce exceptionally large amounts of data, a dedicated server and storage arry for preprocessing and data analysis are essential.
