Autotransporter (AT) biogenesis is a very rapid process, which happens within few minutes from induction of protein expression until presentation of the perfectly folded protein on the bacterial surface. Therefore, the analysis of AT biogenesis demands particular analytical techniques. To this end, we will use a stalled version of the AT intimin. With this tool we can study all the usually transient interactions that take place during biogenesis of the AT in a locked-down state. We will apply the technique of UVinduced site-specific photo-crosslinking to directly identify site-specifically direct interaction partners of the stalled intimin taking place in vivo and to address hypotheses about the roles of specific interaction partners (BamA, SurA).
