Role of regulatory T-cells in the development of donor-specific adaptation after clinical organ transplantation (B01)

Project B1 demonstrated that donor-hyporeactivity is associated with less acute rejections after transplantation in living-related kidney transplantation. These studies will be extended to patients with chronic graft dysfunction after living-related transplantation. In addition, B1 will investigate, if donor-specific hypo responsiveness can be linked to the up-regulation of inhibitory receptors on B-, T- and NK cells. To this end, B1 made the very interesting discovery that a T503C polymorphism in the CD161 gene caused diminished inhibitory capacity in NK cells (collab. B4). The presence of this polymorphism was overrepresented in patients with acute rejection after liver transplantation. In addition, the important work of B1 will also be helpful in defining biomarkers of tolerance, which will ultimately lead to an individualized immunosuppression after organ transplantation.
