Covalent DNA-protein crosslinks (DPCs) are a particular dangerous type of DNA lesions because they interfere with essential DNA transactions. Despite numerous recent insights into DPCs, many crucial questions remain unanswered. Thus, we propose to utilize the analysis of histone-DNA crosslinks as a powerful research paradigm to reveal how cells detect and repair DPCs. This will allow us to combine our experience in DPC research with the chromatin expertise of the CRC to tackle previously inaccessible research questions.
