The goal of this project is to identify and validate genes that negatively regulate antigen-specific T cell function in a cell-intrinsic fashion, aiming at enhancing the activity of low-avidity T cells for adoptive T cell therapy upon perturbation of these targets. On the one hand, we will address the hypothesis that T cell-intrinsic cGAS-STING signaling exerts a negative effect on T cell activity. On the other hand, we will systematically validate a list of candidate genes for their impact on the functional activity of CD8+ T cells and also conduct a genome-wide, unbiased screen to identify negative regulators of T cell activation in a setting of low affinity, antigen-specific activation of T cells.
