This proposal aims at the akquisiton of a high-resolution confocal laser-scanning-microscope, fast acquisition mode, large incubation unit and a module for measurements by fluorescence correlation spectroscopy is applied for. A large incubation unit will ensure physiological conditions for long-term experiments using living cells. The FCS module will allow to measure protein-protein interactions, to perform co-localization studies and to determine intracellular effective diffusion constants. The envisaged instrument additionally covers needs for conventional confocal microscopic measurements that can be performed by point-scanning microscopy and will therefore also contribute to reducing waiting times for the presently available confocal system.The requested microscope will be operated centrally in the technology platform “Cellular Analytics” of the Stuttgart Research Center Systems Biology and is therefore open to all members and associated workgroups across the university. The time of usage for the instrument is calculated as 2600 hours within the first year.
