Contact sites between cellular organelles are crucial places for lipid transport. We previously found the late endosomal / lysosomal sterol transporter STARD3 to be involved in contact sites of lysosomes with the ER, but also with mitochondria. In project A19 N, we will investigate this dual mode of action of STARD3, its regulation as well as consequences in downstream lipid trafficking. Therefore, by investigating the STARD3 switch at a molecular level, we will gain insight into the composition and regulation of contact sites formed by lysosomes and their importance in maintaining cellular lipid homeostasis.
