This project aims at a comprehensive description of thyroid hormone (TH) action during human cortex development using cerebral organoid models. By combining single cell transcriptomics with immunofluorescence analyses, we examine and compare effects of TH deficiency, MCT8 deficiency and perturbed THRA action on glutamatergic neuron lineage differentiation in a cell type-specific manner. Joint profiling of gene expression, chromatin states and THRA-chromatin interactions in single cells and bulk tissue samples is used to dissect specific roles of THRA in gene regulatory networks mediating TH-induced cellular changes.
