ImmunoStroke: From Immune Cells to Stroke Recovery Linking functional immune profiles and ischemic lesion characteristics in human stroke

The first funding period focused on the characterizing effects of stroke lesions and stroke treatment on brain network architecture as well as stroke-related changes in the immune signature and functional properties of peripheral blood immune cells. Building on this recent work we now aim to add lesion network mapping analysis to evaluate the association of changes in the immunome after stroke to alterations in structural and functional brain network architecture. Based on our data indicating substantial changes in the myeloid cell compartment, in the second funding period we aim to further investigate the functional changes in monocytes after stroke. We will focus on different monocyte subpopulations and evaluate changes in function, metabolism and purinergic signaling at the single cell level using flow cytometry, transcriptomics and SCENITH technology.
