The caseinolytic protease Clp plays a central role in protein homeostasis and regulatory proteolysis in bacteria and mitochondria and is a target for antibacterial, anti-virulence, and antitumor therapy. We investigate two classes of modulators of the proteolytic core ClpP of the complex protease. ADEP antibiotics activate ClpP and lead to uncontrolled protein digestion. Here, we explore the altered substrate spectrum. For a new natural product class inhibiting ClpP, we investigate the mechanism of producer self-protection.
