This project will focus on the risk factor Pnpla3I148M and gatekeepers of bile acids (BA) metabolism such as FXR for chronic liver disease (CLD) progression within the gut-liver axis. Specifically, we will study the role of Pnpla3I148M during cholestasis and gut dysbiosis, but also the relevance of FXR signaling for BA metabolism and progression of CLD as well as immune activation. By using animal models and clinical samples, we aim to directly proof our findings towards translation by asking for their relevance as novel biomarkers and as a basis for therapy.
