In this proposal, we will use quantitative high-resolution mass spectrometry to identify proteins re-cruited to a range of defined DNA lesions, such as 8-oxo-dG, cyclobutane pyrimidine dimers and intrastrand cisplatin-DNA adducts, in different cell types and species. Putative new candidates will be further characterised by established biochemical and cell biology techniques. With this approach, we aim to identify complex networks of proteins involved in repair of these DNA lesions and uncover similarities and differences in DNA damage repair in organisms ranging from yeast to humans.
