Project A04 will (i) elucidate how the endocardium promotes cardiomyocyte dedifferentiation and proliferation during zebrafish heart regeneration, (ii) investigate how cardiac endothelial cells induce mammalian cardiomyocyte cytokinesis defects, and hence polyploidization, during early postnatal development, and (iii) identify factors secreted by or on the surface of endothelial cells critical for these processes and manipulate their function to promote cardiac repair in mammals. These studies will provide mechanistic insight into the paracrine control of myocardial regeneration, which will be of high scientific relevance and translational value to cardiac repair research.
