This project focuses on the characterization of inflammation-targeting nanocarriers (NCs) to evaluate their potential for an anti-inflammatory therapy at the oral mucosa. The human oral mucosa may exhibit various pathologic conditions, such as inflammatory or autoimmune diseases. Commonly, anti-microbial or anti-inflammatory topical drugs are applied for the treatment. However, the use of topical drugs at the oral mucosa is limited owing to physical irritation and dilution effects by biological fluids that hinder the adherence of commonly used drug formulations. Therefore, the effectiveness and bioavailability for oral topical drugs is often weak, and patients may suffer from the disease over years, if not life-long.The main objective of this proposal is to improve the bioavailability of topical drugs that are loaded to NCs. This project aims on (1) the characterization of selected NC in intact excised oral mucosa and appropriate 2 D cell culture models which essentially includes determinations of penetration dynamics, of sides effects, and the influence of the NCs of the chemical and the physical barrier of the oral mucosa. (2) the evaluation of effects of anti-inflammatory NCs and drug-loaded NCs in different inflammatory 2D- and 3D- models of the oral mucosa.Here, we propose that newly developed mucoadhesive NCs possess the potential to increase efficacy and bioavailability of topical drugs in the oral cavity and exhibit an anti-inflammatory potential. The obtained results will help to develop novel innovative strategies for prevention and topical treatment of inflammatory diseases of the oral mucosa.
