We have shown that platelets and immune cells jointly drive penumbral tissue loss and ischaemia/reperfusion injury under and after middle cerebral artery occlusion in mice, and confirmed the concept of thrombo-inflammation in the human system using local pial blood samples obtained directly from the cerebral collateral circulation. As next steps we will characterise the contribution of platelet-derived DAMPs and platelet-dependent neutrophil effects on stroke development and collateral blood flow in a tandem experimental/human approach.
