RAS family members are the most common human proto-oncogenes. Analysing intestinal cancer evolution, we found that Kras-mutated alleles are very frequently being amplified at the adenoma to carcinoma transition. We uncovered that this oncogenic dosage amplification induces strong immune signalling programs in intestinal epithelia, including defined chemokine, cytokine and growth factor responses. Here, we propose to (i) study the molecular mechanisms by which mutant KRAS activates distinct immune signalling components and (ii) to characterize the precise nature and biological effects of such Ras-dosage dependent immune signalling programs in oncogenesis.
