We have generated the first large animal model of PDAC in pigs to translate advances in basic research into benefits for patients. It is based on unique pig lines with Cre activatable mutations in the endogenous KRAS or TP53 genes and PTF1-Cre driver lines. After crossing the Cre pig lines with latent KRASG12D and TP53R167H (KPC) pigs metastatic PDAC was induced in young animals. The model will now be validated by cross-species comparison, then further refined by CRISPR/Cas9 based subtype modelling. Finally it will be used to validate and refine novel imaging and therapy approaches, e.g radiation and chemotherapeutic interventions developed within the consortium.
