In the second funding period, we want to further develop the newly produced MCR candidates and improve their selectivity. In addition, we want to use these MCRs on neuronal progenitor cells to quantitatively determine the dynamic changes of the O-GlcNAcylomes at defined time points during differentiation. Similarly, changes in O-GlcNAcylation patterns will be detected in murine stem cells in which critical proteins have been knocked down. We also plan to develop synthetic and chemoenzymatic approaches to UDP-GlcNAc analogs of our MCRs to enable their use in complementary chemoenzymatic labeling methods.
