Funding is requested for a custom-made dual optical parametric oscillator (OPO) laser system for photoacoustic (PA) imaging and spectroscopy using pump-probe excitation. In contrast to conventional OPO lasers, this system will provide independently controlled, wavelength-tuneable excitation across the visible and near-infrared wavelength region, variable time delays (from 1 ns to 1000 ns) between the pulses, fast (pulse-to-pulse) wavelength tuning, high pulse energies (> 70 mJ) and high repetition rates (200 Hz). This will not only enable the development, comprehensive evaluation and further refinement of novel contrast agents and genetically expressed reporter proteins for PA imaging in vitro but will also provide the basis for the translation of this technology to in vivo applications in preclinical and, in the longer term, clinical studies. A dual-OPO laser will overcome the limitations imposed by the OPO laser systems currently available to our group.
