Lipid signalling is commonly understood to comprise all events that involve binding of a signalling lipid to a protein effector, but the mechanisms ensure specific activation of individual effector proteins are poorly understood. Here, we will test the hypothesis that chemical differences between lipid species cause changes in lipid dynamics which in turn constitute a regulatory mechanism for effector protein recruitment. We will use a combination of caged lipids and lipid biosensors to set up an assay for measuring the dynamics of native lipid species in living cells. Our approach will yield novel insights into the molecular regulation of lipid signalling events.
