In order to prepare specimen suited for high-resolution cryo-ET, additional instruments are required. The requested correlative cryo-light microscope (cryo-LM) shall facilitate cryo-fluorescence imaging in a correlated manner with electron microscopy on the 300 kV cryo-TEM for tomography and FIB-milling / scanning electron microscopy on the dual beam cryo-FIBSEM instrument. The main purpose of the cryo-LM is thus to guide the selection of targets for cryo-ET via fluorescent labels (e.g. genetically encoded GFP). Furthermore, the instrument will be used for correlative light/electron microscopic (CLEM) applications . The instrument shall be integrated into the Charité EM core facility to facilitate the cryo-ET workflow, but at the same time provide an interface to the Charité Advanced Medical Bioimaging Core Facility (AMBIO).
