For the elucidation of the mechanism of biological processes the molecular characterization of tissue samples is of great interest. Besides a qualitative and quantitative description of involved molecules the knowledge about spatial distribution of these biomolecules is essential for our understanding of complex cellular organisation and dynamics. One technique that has emerged for this purpose is imaging mass spectrometry as a label-free technique without prior knowledge of the analyte. The Heinrich-Heine University of Düsseldorf (HHU) does not possess a mass spectrometer with imaging capabilities. The use of a mass spectrometer for a high-resolution (5 μm) MALDI-imaging with MS/MS capabilities for the detailed characterization of the analyte is required for the following projects at the HHU: Investigation of the fine structure of the plant cell walls of various cells and parts thereof by cell-specific oligosaccharide mass profiling (COLIMP) and the use of this technique to investigate a plethora of cell wall mutants and specialized cells e.g. those involved in C4 photosynthesis. The cell specific determination of specific C4 protein complexes by their peptide profiles and lipid and sterol profiles. The dynamics and posttranslational modifications of stem cell signalling molecules such as CLE.
