In project A05 the mechanism of substrate recognition by the palmitoyl transferases of the DHHC enzyme family will be investigated by cross-linking mass spectrometry, NMR spectroscopy and electron microscopy. The functional consequences of stimulation-dependent palmitoylation in T cells will be analyzed for selected substrate proteins with regard to Ca2+ signaling, migration and adhesion. Furthermore, the hypothesis that palmitoylation switches are influenced by changes in the cellular acyl-CoA pool will be probed experimentally.
