Our previous data defined a conserved genetic network that specifically coordinates progressive right ventricular (RV) failure in rats and humans. Based on these insights we will investigate additional mechanisms of spatio-temporal control of cardiac gene expression patterns by (i) multiplex RNA-FISH, spatial RNAseq and 3D reconstruction, (ii) characterization of nascent secretomes and (iii) identification of regulated phospho-proteins. At the functional level, we will characterize and phenotype RV signature genes (Penk, Tceal7, Cox4i2, fibrosis-related genes) by AAV9-mediated gene transfer in pulmonary artery banding (PAB) of rats or cardiomyocyte-specific knockout in PAB mice.
