The subclone with the most adverse characteristics determines prognosis of the patient. To identify and characterize adverse subclones, we use the individualized patient-derived xenograft (PDX) mouse model of acute myeloid leukemia (AML) complemented with genetic engineering. In the current funding period, we applied genetic barcoding and described adverse subclones with aggressive behavior upon spontaneous tumor growth. In the upcoming funding period, we will concentrate on adverse subclones appearing upon selection pressure by treatment.
