Synthetic genetic analysis, automated microscopy and image analysis (Z03)

For all projects using yeast as a model organism (P04 - Schwappach, P06 - Thumm and P13 - Rehling) this technical project will provide tailor-made libraries of mutant yeast strains using synthetic genetic array (SGA) technology. These libraries will allow the individual projects to drive their hypothesis-based research program. Systematic analysis will be an asset to the biological problems of the CRC because our insight into compartmental gate function is limited by the extensive use of very few model cargo proteins. In the area of compartmental contact sites such approaches have recently spurred major advances and will continue to do so. The central platform provides a combination of array pinning robot, liquid handling robot, automated microscope, and image analysis pipeline ideally suited to perform systematic localization screens in yeast. Furthermore, the platform will support P07 (Kehlenbach) and P10 (Brose) to implement quantitative imaging assays in mammalian cells with substantially increased throughput as compared to manual microscopy and image analysis. This central project uses existing cutting-edge equipment to provide powerful reagents and methodology to the CRC.
